You may have a trigger conflict with something in System Preferences.
Open System Preferences, go to Keyboard & Mouse, click on the Keyboard
Shortcuts tab, and look for anything that uses ⌘Space. By default, the
Spotlight menu is set to use this trigger.

You can also try changing the QS activation trigger to something else
to see if that works better for you. Double clicking on the
application icon in Applications when QS is already running will
activate it and bring up its command window. Then you can press ⌘, to
go into QS's preferences and modify the activation trigger as
described in Howard Melman's unofficial user guide (see the thread at
the top of the forum).
