Just the obvious, set LogLevel to debug in /etc/cups/cupsd.conf and then
/etc/rc.d/init.d/cups restart
try to print something and see what it says in /var/log/cups/error.log
I always build gtk+ before cups and printing works fine for me.
Do you use foomatic filters?
