Job Opening – Research Scientist Crystal Growth Medical Structural Genomics of Pathogenic Protozoa (MSGPP) University of Washington, Seattle
The successful candidate will become a member of the unit within the multi-disciplinary Medical Structural Genomics of Pathogenic Protozoa (MSGPP) Project which focuses on the growth of high quality protein crystals for X-ray structure determination by high-throughput methods. The proteins are derived from organisms responsible for major global diseases such as malaria, sleeping sickness, leishmaniasis, and others. Characterization and purity of protein samples will be assessed by several biological and/or biochemical techniques to be followed by carefully planned small scale crystallization experiments using pipetting as well as robotic techniques. Great care in preparation and execution of experiments is required. Excellent tracking of results with spreadsheets and databases will be crucial. Participation in development for new protocols and new tools for protein crystallization will be an important component for this position. Special emphasis will be given to co-crystallization of proteins in complex with Ligands to enhance insight in small molecule-binding properties of the drug target proteins under investigation. Specific responsibilities include: - performing protein characterization with a variety of methods including - Limited proteolysis experiments - Dynamic light scattering studies - Determination of purity with gel electrophoresis techniques - Protein crystallization experiments - Programming robots to prepare solutions and set up crystallization experiments -.Preparing stock solutions - Performing the crystallization by robotics - Performing follow up crystallization experiments manually - Analyzing crystallization results - Preparing screening overviews When needed, protein samples will be further purified or modified as e.g. by reductive lysine methylation or other procedures.
