Hi all,
I'm trying to cocrystallize a GTPase with a GTP analog. I tried GMPPNP, but
can only get crystals of the nucleotide-free form. I am now trying aluminum
fluoride with GDP. I have a few questions:
1) Does the active site promote formation of either AlF3 or AlF4-, or is it
more related to the crystalliztion solution?
2) Almost all papers I have seen use either a mixture of AlCl3 and NaF, or
Al(NO3)3 and NaF to form aluminum fluoride. Why don't they use anhydrous
AlF3? Is it because AlF3 in an aqueous medium doesn't ionize easily, and
that NaF with Al-X allows an equilibrium of both types to be formed?
I'm sure they are obvious questions, but my inorganic backgroud is not so
great.
