In the instance where we grew crystals is isopropanol, we found they also grew in PEG6000. The best crystals grew in isopropanol, so we just harvested the crystals into a similar PEG6000 solution. I would imagine that you can get the crystals to grow in something beside isopropanol especially now that you have some crystals to use for sees. You might try t-butanol and other organic precipitants that are less volatile. Or if isopropanol is special, maybe you just need it as an additive at 5% (v/v) or less and can use something else as the precipitant.
I will certainly guess that your crystallization trials are not over yet. :) Jim _____ From: CCP4 bulletin board [mailto:[email protected]] On Behalf Of Chris Meier Sent: Friday, April 09, 2010 3:39 AM To: [email protected] Subject: [ccp4bb] freezing crystals grown in isopropanol condition Dear all, I have a protein which crystallizes in 25% isopropanol, at pH4.5. Does anyone have experience freezing crystals grown in such a condition? What cryoprotectants should I try? Can isopropanol itself act as a cryoprotectant? Any suggestions on how to deal with isopropanol evaporation during mounting? Many thanks and best wishes, Chris
