Yes, you can build an instance of Model-Glue without using XML - you'd create an instance of ModelGlue.cfc and add your controllers and events programattically. For that matter, you could create your own "custom" loader and use it to load ModelGlue.cfc instead of the default XMLModelGlueLoader.cfc. I went this route when I first started Model-Glue and it was called "Call and Answer," because I was used to scripting together events and listeners from Flash.
