I can confirm it happens with Cherokee 1.0.5 and later. I just tried
(1.0).20, .10, .7, .5 and all of them gave me the same error.
Reinstalling 1.0.4 solves it right away, with the same config file of
course.

I can see '  - NEW: Much better UTF-8 encoding" in 1.0.5's changelog. Might
it be related?

As for your test, I'm not sure what it means but 'cherokee -r /tmp' works
and gives me my /tmp listing using both 1.0.4 and 1.0.20, not problems
what-so-ever.
