This usually depends on your topology and the stability of your network. Is there any rhyme or reason to the layout? Is this hub-and-spoke? Is it already laid out in a fashion that would facilitate breaking it up into areas if you wanted to?
Either protocol should work just fine with only 50 routers. You may even be able to use OSPF with a single area if your network is pretty stable. I'd probably divide it into areas anyway if your topology allows you to do so in a way that makes some rational sense. Which protocol do you feel more comfortable with? The configuration of eigrp is simpler, but basic ospf isn't very difficult either.
