This is why I like my fuel pressure gauge. There are several things that can 
stop the flow of fuel and only some of them show up on a vacuum gauge.

Our engine stuttered and quit a while back, of course on a windless day right 
in the major ferry lane...
I finally tracked it down to a loose bleed screw on the Racor. It sounds like 
your problem may be either air in the line, or gunk in the tank fouling the 
pickup tube. I have an outboard primer bulb in the fuel line just before the 
Racor, so pressurizing it is easy, little bubbles coming out somewhere is a 
clue; the bulb also allows a really fast reprime of the engine. We were under 
way again in 30 minutes.
Alternatively, it might be worthwhile pulling the pickup tube and seeing if the 
filter on the end is grotty.
