I'm not sure you're using maildir format here, as you're lacking the trailing slash when defininig your mail boxes:
MAILDIR=$HOME/Maildir/ DEFAULT=$MAILDIR
:0: * ^X-Spam-Status: Yes .Spam/
I'd say you're using mailbox format in maildir-named directories.
What can I say... the configuration works for Maildir not mailbox. Procmail automatically puts a "/" at the end of Maildir and if you then append that to the "Default" you would get a double slash in the path. Just watch the logs and you will see.
