Yes, I weighed it down that way yesterday. I put it in paper towels and put the paper towels between two plates and then put a couple things on top. Much better than trying to squeeze the water out by hand, which is also likely to break up the tofu anyway. All this good preparation didn't keep me from pretty much ruining dinner. But that's another story.

Let it sit for at least a half hour. This will definitely help drain the water. It's a pain to squeeze tofu. At least for me, weighing it down gets more water out.
