The difference is that with greylisting, you keep track of the know good senders. So only the first e-mail to/from a particular person coming through a particular mailserver is delayed.
The "comeagain" filter keeps a database of sender/recipient pairs it's seen before. It only disposes of pairs if no mail is exchanged for 30 days. That should keep delays to a minimum.
