THE PINEAL GLAND - MELATONIN.
The pineal gland is a small structure, shaped like a pine cone, situated
between the cerebral hemispheres of the mammalian brain. Until recent
times, the actions of the organ were little understood. Isolation and
characterisation of melatonin from the pineal initiated a new era since the
compound was found to affect skin pigmentation in amphibia and to inhibit
the development of gonadal function in young animals as well as the action
of gonadotrophins in mature animals [page 1121]
Melatonin reverses the darkening effect of MSH on the skin of amphibia by
stimulating aggregation rather than dispersal of melanin granules within
melanocytes, causing lightening of skin colour.
Melatonin synthesis occurs from tryptophan via hydroxylation and
decarboxylation to form serotonin [page 672]. Acetylation and methylation
of seotonin yields melatonin.
This synthesis is controlled by epinephrine via the stimulatory action of
the latter on adenylate cyclase of pineal cells and a subsequent
stimulation of protein kinase.
