I was looking at the header of a suspected spam email, and it didn't have
any declude headers added. I thought this very strange so I looked at the
declude spam logs and found that the email had generated this error. Looks
like it sailed right through without getting looked at. Any idea why it
couldn't lock it? I checked the disk, plenty of space, spam folder is OK
and a lot of other spam is getting caught.

This will occur if the Q*.SMD file is missing. This can happen if an on-access virus scanner deletes the file (which would be unusual for a Q*.SMD file, though), or it could also happen with some versions of Declude JunkMail if Declude Hijack blocked the E-mail.

The "error #32" indicates a sharing violation -- meaning that some program has the Q*.SMD file locked, even though it should not normally be accessing it (it's possible that Declude could be doing that). What version of Declude are you running (you can type "\IMail\Declude -diag" from a command prompt, exactly like shown, to see the version)?
