. ./set-mapnik-env
set | grep MAPNIK
resulted in more or less what you've mentioned, but without:
MAPNIK_DBPASS=
MAPNIK_PREFIX=
these are not mentioned in set-mapnik-env
BUT
./customize-mapnik-map
and also
./customize-mapnik-map>osm.xml
resulted in:
bash: ./customize-mapnik-map: /usr/bin/perl: bad interpreter: No such
file of directory
Any ideas?

prints a lot ("osm.xml") using the "osm-template.xml"
(later save this to the file with appending "> osm.xml")

customize-mapnik-map just exchanges parameters in this
file and "print out" osm.xml. (">" redirects it to a file..)

I couldn't find "inc" subfolder in mapnik folder. There's "include"
folder though with hundred of hpp files.
