We develop Affinity Diagrams in an FDD manner with all stakeholders in the
room at once, (very tedious and I'm not sure this is the best use of
everyones time), but it does get everyone's input.

Now testing these diagrams, what would you test them against? Requirements?
Prototypes? Data sets?

Like Todd's info post we rely on different stakeholderrs input upfront. We
do revise the diagrams after protyping if inconsistencies come up, (pretty
rare as AD's are more the "What" then the "How").

Possibly your question could be rephrased as is there any way to test
requirements, and from an interface perrspective this could be done via a
prototype.
