The only way to make sure that your email will be received - not junk-
listed and delivered in a reasonable timeframe - is to partner with a
reliable third-party email provider that uses industry connections and
ISP whitelisting to prevent spam blocking.
We are launching a members-only site with a confirmed email addy as
the membership requirement, and a 3rd party provider was the only way
to reliably do this.

Ram, no disrespect to you, but any method that would reliably help
you get your legitimate emails past junk mail filters -- WITHOUT
cooperative action from the intended recipient -- would also be an
excellent method for constructing more effective spam.
Let's not go there, please.
I have seen several sites that finish the email signup process by
either 1) displaying a request that the user add the sender's email
address to their personal address book (to avoid 'unknown sender'
fileters) or 2) asking the user to look in their Junk folder for an
email that has a specific title.
While these methods are somewhat awkward, they generally do not
impose a heavy burden on the user or the copywriter for your email.
