Some of my beta testers use very large libraries even with a ReadyNAS, although it's a pain. Your HW definitely sounds powerful enough. There's a few things to consider WRT iPeng:
1. iPeng caches long lists. This helps a lot with browsing after it has done so but can slow things down during the process. It does so after each change to your library only. 2. With this size of a library you definitely want to set "Prevent Sleep Mode" or "keep Connection" in Settings->iPeng settings at least until the caching is done (for convenience use "keep connection"). You can see caching is done if the blue progress indicator in the "Albums" view finishes. 3. Make sure you don't have a virus scanner interfere with your index files. That's a known issue especially with McAfee. Time is pretty much exclusively determined by server performance. 7.5.0 embedded beta will speed things up a LOT wrt these queries.
