We have a project that is using Django on the backend.  We are connecting to it via PyAMF, which has proven to be pretty solid.  As far as Django goes, it is probably the most popular Python framework out there.  As Flex developers, we are insulated from having to know the innards of the Python/Django, as we have a dedicated backend team of Python developers.   If you have specific questions re: Python, Django or PyAMF. shoot them over.
