Vacuum leak at the manifold would cause idle issues (revving, etc.) but most likely not a misfire.
OBD-II codes will tell you which cylinder is misfiring. >From there you can try things like replacing a plug wire or plug (cheapest things first) to narrow it down. If that solves it, then be sure to change all plugs & wires so that they have the same age. If you can get hold of ONE to test, though, it may be cheapest or free, in case that doesn't solve the issue.
