I recently looked into this for a client. The good news is that it's
possible. See my comment at the bottom of:

Apart from the things you need to do on the DSpace side, you also need to
touch base with the handle.net folks at CNRI, to redirect the resolution of
the existing handle prefix to your DSpace server.

Things that are not possible to my knowledge:
- Resolve SOME handles for a particular prefix without resolving ALL of
them. When you make the switch at CNRI, the other third party IR won't be
resolving the other handles anymore.
- Register handles for *different* prefixes (new items). While the usecase
of "keep my old handles and their old prefix working after a migration"
works fine, the new repository will only register/mint handles for one
particular prefix. It might be a nice customization/addition in the future
to have different handle prefixes for different collections/communities,
both for existing items as for new ones.
