This is definitely doable. The default boot disk seems to be the Duo, but
if you change the settings in the StartUp Disk control panel it should boot
fine off the Dock's hard drive if you have a System Folder installed.
I find it useful for optimizing the Duo's drive once a week with Plus
Optimizer.
