I have done similar for my portable antennas.

I loaded the "Coghlans-Camp-Laundry-Reel" ($6) with 24ga teflon (what it
would take) ended with one at 75+ feet and the second with 100+ feet. I
used yellow and red wire. I like that teflon has silver plated wire and
it has lasted many-many years of use. I added banana plugs at the end,
too. I have tried chalk lines, but I go back the the simple/cheap one...
I had "free" access to wire at one time.

I looped the wire and taped it with electrical tape.
For winding, I use the SOTAbeams midi winders. They are the same
length as my KX3. I tried camping clothesline reels, but they would
bind and it just took too danged long to wind them up.

For rope into trees, arborist throw line is great. It is lightweight
and specifically designed to not get stuck in trees.
