Do you mean that while you're at work, you want to send all mail through
your work account?  Yes, you could manually set the work account as default
in Tools -> Accounts daily.  It might also be possible to do using
AppleScript.

Or, perhaps a better option, you could use SMTP Location X to change your
SMTP server settings for all of your accounts depending on where you are.
That way you can send from whichever account you want wherever you are.  You
can get SMTP Location X from ScriptBuilders @ MacScripter.

Because I don't have a .Mac mail account, I don't know whether their SMTP
server will relay mail from any address or just from an @mac.com return
address.  You could give it a try.  Make sure that you set the
authentication settings properly in your work and home accounts, using the
"SMTP Server Requires Authentication: Log in Using..." option.  But it might
be to your benefit to try the SMTP Location X script first.
