Do you mean that while you're at work, you want to send all mail through your work account? Yes, you could manually set the work account as default in Tools -> Accounts daily. It might also be possible to do using AppleScript. Or, perhaps a better option, you could use SMTP Location X to change your SMTP server settings for all of your accounts depending on where you are. That way you can send from whichever account you want wherever you are. Because I don't have a .Mac mail account, I don't know whether their SMTP server will relay mail from any address or just from an @mac.com return address. You could give it a try. Make sure that you set the authentication settings properly in your work and home accounts, using the "SMTP Server Requires Authentication: Log in Using..." option.
