Mr. Bill's advice is good.  If you only have one
flash, then you're better off shooting in daylight,
but a cloudy or at least hazy day will be needed to
prevent glare and hotspots.

Why includethe background in the photo at all?  Adjust
the framing so that you only shoot the painting.

Autofocus and AV are completely separate.  AF should
work fine on this.  Just make sure your camera back is
parallel to the painting.

As Mr. Bill suggested, an incidient light meter would
be best.  Assuming you don't have one, however,
acquire an 18% standard gray card (available from good
photo stores where you are, I hope), and meter off of
that.  Then manually set the exposure on your camera,
and bracket.

A couple of other thoughts:  It sounds like you're
going for prints as the end product.  Be aware that an
automated processor is NOT going to get good prints
from these images.  An auto procesor will assume that
the print is, overall, 18% gray, so your white
background will come out gray.  You're goingn to need
a decent photo lab to make good prints of these images.
