The modem is locked with a /var/lock/LCK..MODEM file.  That keeps other
users from accessing the device when it is being used.  Just delete it (as
root) and continue on with your mission.  It might also be that you are
trying to run multiple instances of minicom, so check that with `ps aux |
grep minicom`.  I think there's even a nolock option in minicom, but that's
not a good idea generally.  Also in X, try xminicom and you get the same
interface in a shell.
