I've only done it once, and even then, it was with quite a bit of trepidation. The server was hung so bad it was literally taking minutes just to process a single command. Ended up having to do a hard boot (ouch!). IIRC, the problem ended up being the version of Symantec AV for Exchange (possibly coupled with SAV Corp ed). Since upgrading to Symantec Mail Security, we haven't had any issues such as that. For several months we were't running any AV on Exchange (all inbound/outbound was handled by a separate gateway). Boy, did that leave a bitter taste in my mouth of Symantec products. If we didn't have an Enterprise agreement with them, I'd be seriously pushing for Trend or Sybari.

I have never tried killing Store as I haver always been to worried it may corrupt the DB, so it's ok to kill Store.exe ? In perfmon, I have been watching Current Disk Queue Length, it has a maximum of 1 so I think that is ok. I have been running with out GFI MailSecurity running for the last 48hrs and the problem has not occurred. I just enabled the VS API and the RPC Averaged Latency shot up to 30ms and kept climbing. Disabling VS API, it gradually came down to <10ms. Time to call GFI again methinks....
