It's possible in both 5.5 and E2K3, though done slightly differently....
and to facilitate the move to E2K3 I'll describe the setup for 5.5
slightly differently.

In the 5.5 Admin remove the mailbox which is currently assigned as the
primary NT account from the box on the General(?) tab. Assign that
account and the other user accounts as mailbox owners on the permissions
tab. 

When your ADC is put in place it will replicate the mailbox object over
as a disabled mailbox enabled user with the same access permissions.
