Do you have any monitoring software running, what happens if you stop
it?
Do you have any antivirus running, what happens if you disable it during
that time?
Check what processes are eating the most CPU and memory when the hangs
happen. 
Also check disk activity during that time, anything that is taking up
more than normal?

If you have users on Outlook 2003, there are some hotfixes you should
apply to your 5.5 server. Look on MS's  web site for details.
