I don't see why not. I've done the same thing with Exchange 2003 running on Win2k3 R2, live during the middle of the day, with no isues.
I suppose if you're crossing the 2tb barrier, it would probably make a difference, but that would require major surgery anyway, unless you had configured it as a GPT disk in the first place (and doesn't 2008 R2 do that by default? I can't remember off the top of my head).
