The expectation is that xvfb-run is sufficient to do whatever it has to do to get itself working. If that means triggering the launch of Xquartz, then it needs to do that, and I'm also surprised that running Xvfb would not trigger it. The xvfb-run script is just a wrapper around Xvfb itself that handles xauth, avoids race conditions when picking various arbitrary/dummy screen parameters, and graceful shutdown. Probably the simplest (or at least first) test would be to see if just "Xvfb :10" from the commandline fails.
