I ran into this just last night when I moved my external hard drive with my iTunes library on it to another machine...
Assuming you have admin rights on your machine, open Windows Explorer and browse to My Computer. Right-click the drive in question and choose Properties. Click the Security tab. Change the permissions for the account you log in as to Full Control. You should then be able to use the drive with no issue.
