I did a test with this by creating a hotspot in FrameMaker and saving the document to PDF. The hotspot is converted to an Acrobat Link object. As far as I can see, Acrobat Link objects don't have a tooltip property. You can see this by creating a Link by hand with the Link tool. I don't see any place in the Link properties dialog box where you could specify a tooltip. So I am not sure how this could work going from Frame to PDF.
