You should be able to do that, except that the stick needs to be
inserted before you boot Freedos. You cannot "hot swap". If there is
no USB stick when Freedos starts, then there is no drive letter, and
it will not detect a stick you plug in later.

("Stick" can also be external drive, external floppy etc. Then you
will need to change in the BIOS perhaps, or maybe the BIOS setting is
only the boot order like I believe you suggested at some point.)
