I'm not sure what you mean by, "determine the font size of a given variable not flow." Generally, the "variable" is a field value from your data, and does not have any inherent font size. When you call out the variable in a text flow, that's when you specify what font face and point size you want to set it in.
If you're asking about how to determine what the actual output size of a particular bit of text would be after copyfitting, then again, I first need to know which kind of copyfitting you're talking about, then I might be able to tell you whether it's possible.
