No. Here's a quote from the PlusOptimizer 1.3 Read Me, dated 8 January 2001. (This is the version distributed on the DiskWarrior 2.1.1 CD.)
"You cannot optimize a disk currently in use by Mac OS X. To optimize a Mac OS X disk you will need to be started from a Mac OS 9 disk." So, you can't run PlusOptimizer from Classic, but you can by booting off the DW CD, which is the best way to run disk optimization software, regardless of brand or operating system.
