Can you turn off all the firewall functions on the router, to make sure the problem isn't Mail.
What happens if you look in the system or console logs or watch the Activity window in Mail while you try to connect?
I don't know what protocol you're using, but I'm connecting to GMail in IMAP mode and incoming needs port 993 open and outgoing is set to 'use default ports 25, 465, 587'.
