Is there any advantage to a USB/FW400/FW800 and eSATA on an enclosure with an optical drive in it. Can the drive run as fast or faster than USB?

Well, that would, of course, depend on the drive.

Here's one that OWC recommends for Mac Pro -- just to take an example.

Pioneer DVR-218L 22X DVD Burner

Its internal interface is SATA. I would venture it's SATA I, which is quite a bit faster than the max r and w speeds (below).

Data Transfer Rate (Data Read - Sustained)
DVD max 22.16 MB/sec
CD max 6.14 MB/sec

Data Transfer Rate (Data Write - Sustained)
DVD max 27.7 MB/sec (22x DVD-R)
CD max 6.14 MB/sec (40x CD-R)

27.7 MB/sec is 221.6 Mbps. That's about 80% of the normal throughput of USB 2 (dedicated).
