For various reasons, I am using all 3 systems, but the only way I knew how to 
set up a layer for GeoWebCache was piping it through GeoServer first. I guess 
that's not the case.

Either way I worked out the problem was to do with GeoServer needing to 
'rediscover' the MapServer layers.
Originally the same map file had been set up for some EPSG:4326 layers when it 
was set up.
At that point the bounds were -180,-90,180,90. Of course, without refreshing 
the settings, GeoServer was only making requests to satisfy the apparent bounds 
in metres rather than degrees.

That meant it was going to be a pretty small bounds!

All good now.

Are you using GeoServer configuring wms cascade for MapServer?

If this is the case, and you are only using it for the integrated GWC, it seems 
to me it is overkill. I would deploy a stand alone installation of GWC to cache 
mapserver layers.

Please forgive me in case I totally misunderstood your problem
