The ESRI UC attendance is a good surrogate. I would figure that about 1 in
10 US GIS professionals attend the UC each year.

The UC is a good surrogate because:

1. Attendees are generally GIS professionals - not just people who use GIS
as part of their profession

2. Even hard-core ESRI users use other software occasionally - especially
Python. And people who normally use other software sometimes attend the UC.

I say 1 in 10 because there are likely some shops that send most of their
GIS professionals and many shops that send almost none.

ESRI claims 12,222 attended in 2008. My estimate yields 122,200 GIS
professionals in the US.
