Assuming they are both configured correctly then yes, the two machines
should communicate directly.

Routers get involved when the destination of a packet appears to be on
a different network segment.  Then the client needs to know the address
of a router (on its own segment) which can handle traffic to/from other
network segments.

Is there a different question perhaps that you're trying to answer?
