I also use Gnucash on a couple of installs of Windows 10 Pro and have
Windows Defender running.  My update/install process is to download the
Gnucash setup.exe, right click on it and display properties and then click
the Unblock checkbox and then simply double click on the .exe file and
click OK when necessary to allow the install or update to proceed.  My file
size on Windows is 141 MB (148,789,209 bytes) and I would expect your
downloaded file to be the same.

I'm with nvsoar on this one and think you need to download the Gnucash
setup.exe file again and make sure the download completes ok, then try the
re-install again.  Either manually uninstall the currently installed
Gnucash or let the new install uninstall the current version for you -
it'll prompt you to uninstall the currently installed version.
