Of course you can. Appengine is a different sort of environment to Ruby on Rails / LAMP stacks (PHP/MySQL). I would say that your learning curve would be quite sharp if you don't know much about programming. There are a fair few concepts to grasp before you can become effective with AppEngine or for that matter LAMP.
AppEngine is definitely the platform if you want your application to scale without you having to worry about running infrastructure. I would recommend working your way through the initial tutorials and see if it suits your level of skill. Its difficult to say yes or no if you dont have any experience in writing code.
