You have already done some research, and presumably found nothing which will help with (2). One reason for that is that the decision about what to include is rather subjective: what is a "major" city as opposed to any other city? What do you class as on the route? Is a city with a bypass (so that the route actually goes round it rather than through it) classed as on the route for this purpose? I can't think of a straightfoward way of doing it.
