Notification Center doesn't allow for anything other than the app icon. The app store screenshot is showing using a custom Growl display (i.e. GrowlTunes is talking to Growl).
Unfortunately I didn't think to change the screenshot until after GrowlTunes was approved, it does look somewhat like NC. But that is what the note about being able to optionally use Growl is about.
