Sounds like you have some sort of system problem. I've tested extensively on ubuntu and i've never seen this before.
If you run tail -f /var/log/messages do you see any errors? Do you see any errors about hplip on boot? HPLIP should be started on boot up. As a work around you may be able to add /etc/init.d/hplip start to the System -> Preferences -> Sessions - > Startup Programs But HPLIP should be running at boot.
