I am curious, is there much software that will use partitions?
Not that I'm aware of; I'm only familiar with non-commercial goodies.
What do they buy you over multiple TN3270 sessions?
Pop-up windows, screen partitioning, etc., and a single logon. Or anything else you can think off. Do you ever need to look at 3800 print data (e.g., a wide dump?) - set your partition to 20*204 and look at the text without wrapping (with supporting software). Or you need to debug software written for a 12*40 screen size? When you get sick of 24*80 screens, set yours to 80*24 <g>
The only 3290's we ever had were used as consoles, so I never had a chance to play with this. I think ISPF supported them at one time (maybe still does).
The nice thing is that you don't need a real 3290 (although I have one to play with); most of the quality emulators do provide 3290 and graphics support, with color as a bonus.
