I use ISPF EDIT on UNIX files which have records much greater than 255 bytes. And I just created a VB/6000 data set, added a record with a | in column 1000, and saved it just fine. I then went into edit on it again and all was well. This is on z/OS 1.12. The old TSO line editor (EDIT) doesn't like this data set.
