OK, well after a little more fiddling, I've got it working again. 
Here is what I did: 
1) First I created a new user account but this time through the gnome menus 
(Administration->Users & Groups), and made sure that 'Use audio devices' was 
checked on the 'User privileges' tab in the settings. 
2)Then I logged into this account and found that the sound worked. So then I 
copied the .gstreamer-0.10/registry.i486.xml file from this account into my old 
account. Now audio works fine in both accounts.

I'm not sure why the audio didn't work the first time I created a new account, 
I would have thought it would have enabled audio by default.
