It sounds to me like your Mail directory (~/Library/Mail) has become corrupted. Next thing to try, if you haven't already, is to move ~/Library/Mail to the Desktop and relaunch Mail. If that doesn't help, try creating a new user in OS X, login as that user, and set up Mail there..... if you get the same error, that tells you it's either your install of OS X or something wiggy with your ISP; if you can get your email with the other user, then the problem is isolated somewhere within your original folder within /Users/; you'd want to try trashing anything within ~/Library/ that has to do with mail (namely, anything in Preferences that has mail in it's name, and the Mail folder).
